NRTIs work by structurally mimicking the nucleotide building blocks that HIV normally uses to generate copies of viral DNA. Once NRTIs are integrated into the growing HIV DNA chain during transcription, the polymerization of viral DNA is discontinued. The result is a dysfunctional copy of viral DNA and disruption of the viral reproduction process.
