PIs, as their name suggests, work by blocking the activity of proteases. Proteases are the viral enzymes responsible for accurately cleaving newly formed viral proteins that result from translation of viral mRNA. Therefore, PIs prevent the formation of viral proteins, resulting in a paucity of available proteins for the formation of new copies of HIV.
